On a perfectly functioning vbox host, I don't have it either:
galacticdominator% ls /dev/vboxdrv
ls: /dev/vboxdrv: No such file or directory
but I do have:
galacticdominator% ls /dev/vboxdrv*
/dev/vboxdrv0
